Zhuyu Zhenggan Decoction Ameliorated Rat Liver Fibrosis via the Phosphoinositide 3 Kinase-Protein Kinase B Signaling Pathway Based on Network Pharmacology Analysis

Liver fibrosis is an outcome of many chronic liver diseases and often results in cirrhosis, failure even hepatocarcinoma. Zhuyu zhenggan decoction as a classical traditional Chinese medicine formula used to clinical practice while its mechanism unclear. The aim this study was investigate the anti-fibrosis effect explore molecular mechanisms by combining network pharmacology animal experiment. carbon tetrachloride-induced rats were treated with three doses decoction. function evaluated histopathological examination biochemical detection. related protein phosphoinositide 3-kinase, phosphorylated kinase B assessed Western blot. herb-component-target constructed combined pharmacology. Network found that 119 potential active compounds obtained through analysis bioavailability drug-like properties, involving 142 anti-liver targets. Gene ontology enrichment yielded 625 gene entries, most which biological processes functions. Kyoto encyclopedia genes genomes pathway screening 117 signal pathways, including 3 kinase-protein pathway, mitogen-activated tumor necrosis factor etc. Compared model group, can ameliorate pathological damage rat tissues, significantly reduce serum aminotransferase, aspartate malondialdehyde levels increase superoxide dismutase inhibit alpha, interleukin-1 beta interleukin-6 level. blot showed down-regulate expression phosphorylated-phosphoinositide 3-kinase protein. These suggested markedly alleviated regulating anti-oxidation, inhibiting pro-inflammatory factors 3-kinase-protein signaling pathway.
